In general, it depends on the circuit - some are easy, some are difficult. Some problems you might face -
Mechanical
–Finding precise footprints for the components
–Determining appropriate positions for IO connections
–Placement of components to minimize trace length
–Placement of components to avoid overheating
Electrical
–Sucessful routing of traces
–Avoidance of crosstalk
–Proper selection of trace widths for given node
–Avoidance of ground or other loops
